Fast developments in biophysical tools (single molecule, cryo-electron microscopy,…) provide precise measurements on timescales of the recognition and folding processes of biological molecules and of their complexes. Concurrently, the number of crystallographic structures of biological macromolecules is steadily increasing yielding its wealth of atomic data and architectural insights on molecular recognition modules and folds. The presentation will focus on RNA molecules and RNA-protein (RNP) complexes, the study objects of predilection in our unit. The new data show that RNA conformational rearrangements stabilized or induced during the hierarchical RNP assembly process drive cooperatively complex formation through multiple pathways. The analysis of such data raise complex physical questions. Besides, the relevance to biological in vivo situations is not always easy to grasp or to communicate and the resulting experimental difficulties, especially for direct cellular imaging, are constantly raising. Finally, the data integration of the resulting static and dynamic views, together with the vast genomic and structural data, presents new computational challenges.
